Optimization of Culture Media for Miniature Roses In-Vitro Culture

Abstract

This study investigated the effect of benzyladenine (BA) concentration in culture medium for miniature roses in-vitro cultivation. Nodal explants of two rose cultivars (MR01 and MR02) were cultured on Murashige and Skoog (MS) media supplemented with four concentrations of BA (0, 1, 2, and 3 mg/L). Number of shoots, shoot length and number of leaves in initial, first, second, and third sub-culture stages were observed. Results indicate that both cultivars responded differently with MR01 growing well in all sub-culture stages while MR02 only in the first sub-culture stage. MR01 responded to BA concentrations from the first to third subculture stages for all observed variables. Added 3 mg/l BA in culture medium increased number of shoots from 1.6 in initial stage to 2.0, 4.6, and 3.4 in first, second, and third sub-culture stages, respectively. This treatment also increased shoot length and number of leaves of MR01 well in the first to the third sub-culture stages.
